Wcf Genetic Testing Standards

To enable the PKD test to be used in other breeds, our laboratory has set rigorous standards that consider the possibilities of phenocopies, disease heterogeneity, sporadic mutations, "de novo" mutations and breeding practices. In order to transfer a genetic test to a different breed, the following criteria have been established and we attempt to follow these standards before releasing tests for other breeds:

1. Standard clinical signs and diagnoses must first be confirmed. In the case of PKD, proof of cystic kidneys by ultrasound has been the gold standard. For PKD, the cysts need to be multiple in one kidney or present in both kidneys. Once these diagnoses were confirmed, samples were considered as "voucher" specimens for DNA testing.

2. The DNA test had to match the clinical diagnoses.

3. Once an individual was confirmed as positive by clinical and genetic diagnoses, related individuals were collected to confirm the mode of inheritance. This step is taken to help prove if a mutation is a new, sporadic event, or an inherited mutation. New sporadic events pose less risk to the population than an inherited mutation. Proving the mode of inheritance also supports that the disease is caused by the same mutation and potentially inherited from Persians.

4. A variety of cats from different lines need to be examined to support the test for the breed. For British Shorthairs, we were cautious as to whether this mutation was due to outcrossing with Persians, or a "de novo" mutation within the breed. Hence, some cats could have PKD from Persian crosses, while others have a new mutation. Some breeders perform unacceptable outcrosses, thus this step also helps to determine breed risk.

5. Families from new breeds were proven by parentage testing. Known and unknown accidents happen, thus, all research pedigrees are confirmed by parentage testing.

6. Some percentage of the breed must be examined. Hence, again, is this a localized problem, or an inherent problem within the breed. Different breeding practices for different associations and countries need to be considered. Hence, PKD in British Shorthairs from Australia may not imply that all British Shorthairs in other countries are at risk if Persians have not been a common outcross. This percentage is a slippery slope and should ideally represent the effective breeding population. This aspect of the testing may be the most difficult to establish.

7. The breed should have a clearly defined outcrossing program with cats that have a high risk for the disease, such as Persians, Exotic Shorthairs and Himalayans. Parentage testing should be used to confirm this outcrossing.

Carriers are NOT the Enemy

🧬 “This cat is a carrier.”

And suddenly people panic.

Let’s clear this up:

👉 Being a carrier is NORMAL. Having a variety of genes is normal. Mutations are normal and happen all of the time.

In fact, EVERY cat carries something.

A carrier simply means:
The cat has one copy of a gene that is expressed, but carries another that it does NOT express.

For the purposes of this post, we'll consider carriers as those that appear normal, but carry another gene they don't show but can pass on to some of their offspring.

If the gene that is carried (i.e. the recessive gene of the pair) is a "bad" gene, the only time it matters to the health of the kitten is when:
👉 Two carriers of the SAME "bad" gene are bred together

That’s when you might produce affected offspring.

This is where DNA testing shines:
✔️ You can safely breed carriers if they have other attributes valuable to the breeding program
✔️ You can AVOID risky pairings to avoid producing affected kittens, even using carriers
✔️ You don’t have to remove valuable cats from a program right away, but you CAN choose non-carriers from the next generations and trend towards clearing the gene pool over time without removing valuable diversity in the lines and positive traits in those carrier animals. If you are working with carrier animals to preserve certain lines, it is extremely important to use DNA testing as a tool to avoid unwanted pairings and to assist in removing a detrimental gene from the population over time.

What's the effect of eliminating all carriers instantly from the breeding pool (particularly in a small population)?
✔️ Cull all carriers in one go - the positive - you remove the gene entirely right now, and just have to test new breeders to prevent the gene coming back into the program
đźš« That shrinks your gene pool, which can create bigger problems long-term through genetic bottle-necking / inbreeding suppression
đźš« Loss of valuable positive traits in those carrier lines that may be the "baby thrown out with the bathwater".

Recessive Health Gene Considerations

Obviously the choice of removing an animal from the breeding pool due to genetics depends on the trait, the other value of the breeding animal, and the severity of the undesirable effect of the gene. If a large number of equal value breeding cats are available, it makes sense to remove the carriers in the first generation. However, for most breeders, our lines and number of animals is limited. It may cripple an already narrow gene pool to remove the gene entirely in the first generation. In these cases, DNA testing allows a breeder to avoid pairing two animals carrying the detrimental gene, avoiding producing affected animals and allowing for the chance to create and preserve a valuable line kitten that doesn't carry the gene in the future.

Dominant Health Gene Considerations

If a gene is dominant (expressed with only one copy) and particularly if the gene has a seriously undesired effect, it makes sense to remove all animals carrying this gene from the breeding program entirely. If the gene is simply carried without undesired effect, then DNA testing can influence decisions for future breeders, allowing you to w**d out less desirable traits/genes over time / generation(s) without producing affected animals while preserving the valuable lines.

Smart breeding isn’t about fear—it’s about informed decisions. Understanding the inheritance and effect of the genes in the lines is crucial to making intelligent decisions for the overall improvement of the line over time.

What Is a DNA Panel, Really?

So… what exactly is a feline DNA panel?

Think of it like a genetic report card for your cat.

Companies like wcf, fractal, and others analyze your cat’s DNA and give you a right way.

✔️ Traits (coat color, pattern, hair length, etc.)
✔️ Genetic health markers
✔️ Blood type
✔️ Sometimes even breed ancestry markers

It’s not just “fun facts” though—it’s valuable information.

Instead of guessing:
“Does this cat carry long hair?”
“Could this pairing produce X trait?”
"Does this cat carry a known genetic health issue?"

For breeders, that means:
